Project Title- Analysis of dopamine homeostasis at the nucleic acid level using the Drosophila Genetic Reference Panel


Overview of the project-
The purpose of this project is to identify cell-signaling pathways that influence the function of dopamine. Dopamine is a neurotransmitter that acts as a trophic factor for and regulates the excitability of neurons. We observed that among a set of 39 naturally selected populations, there is significant variation in the expression levels of dopamine-related genes. One bioinformatic approach we are using to characterize these differences is expression quantitative trait locus (eQTL) analysis of this microarray dataset. The prediction is that DNA sequence polymorphisms, in genes that are directly and indirectly related to dopamine signaling, influence the expression of dopamine-related genes.

Overview of computational methods-
Managing and analyzing the SNP and microarray datasets requires the DLX cluster. A large memory allocation is needed to generate SAS databases with these data. Those databases can then be used to generate a JMP Genomics database that is used to perform SNP-trait analysis. Each of these methods is currently available at the University of Kentucky.
